Peptide bonds in selank are susceptible to hydrolysis under strongly acidic or basic conditions, and the terminal proline residues are vulnerable to exopeptidase activity in biological samples. Lyophilized powder stored dry at -20 °C typically remains stable for extended periods, whereas aqueous solutions degrade faster and may lose measurable purity within days to weeks depending on pH, temperature, and microbial load. Repeated freeze-thaw cycles promote aggregation and adsorption to container surfaces. For analytical work, solutions are usually prepared fresh, kept cold, and used within a single working day.
Handling follows standard practice for research peptides. Material is weighed in a low-humidity environment because the powder absorbs atmospheric moisture. Purity is reported as the percentage area of the main peak in a chromatogram, with specifications commonly set at 95 percent or higher; values below that threshold indicate the presence of truncated or modified species. Residual trifluoroacetate from purification is often present and may affect mass balance. Certificates of analysis should state the analytical method, the column and gradient used, and the lot-specific retention time so that results can be compared across suppliers.
Published clinical evidence is limited. Most controlled trials were conducted in Russia, enrolled modest numbers of participants, and appeared in Russian-language journals, which restricts independent verification. Reported outcomes include lower anxiety scores, improved attention and memory measures, and changes in fatigue ratings. Reviews written in English note methodological limitations such as small samples and inconsistent endpoints. Whether the compound produces clinically meaningful benefit relative to established anxiolytics is therefore an open question rather than an established finding.
Selank is a synthetic heptapeptide with the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro. It was designed at the Institute of Molecular Genetics of the Russian Academy of Sciences as a structural analogue of tuftsin, a naturally occurring tetrapeptide fragment of the immunoglobulin heavy chain. The added Pro-Gly-Pro tail was intended to slow enzymatic degradation and extend biological activity. In Russia it is registered as an anxiolytic nasal preparation, while regulators elsewhere have not approved it for clinical use.
Proposed mechanisms centre on modulation of the GABAergic system, with reports of altered expression of genes related to GABA-A receptor subunits and changed monoamine turnover. Some studies describe inhibition of enkephalinase, the enzyme that degrades endogenous enkephalins, which may prolong opioid peptide signalling. Effects on brain-derived neurotrophic factor and on cytokine expression have also been reported. These findings come largely from animal models and small human studies, and the precise primary target remains unresolved.

| Storage of solution | 2-8 °C for short periods | Freeze-thaw cycling promotes aggregation and surface adsorption |
Naming conventions place Selank in the same research family as Semax, another Russian-developed peptide investigated for cognitive effects. The two compounds share a lineage but differ in sequence and in the biological systems proposed as their targets. Semax descends from ACTH fragments, whereas Selank descends from tuftsin. Publications sometimes identify Selank by its full peptide sequence or by laboratory codes rather than one uniform trade name. Because replication outside Russia is limited, reports on its properties are best read alongside the study design and the purity of the material tested.
Selank is a synthetic heptapeptide developed in Russia during the 1990s. Researchers at the Institute of Molecular Genetics of the Russian Academy of Sciences designed it as a stabilized analog of tuftsin, a naturally occurring immunomodulatory tetrapeptide. The compound has been studied primarily for its reported anxiolytic and nootropic effects. It remains largely unknown in Western pharmacology and is not approved as a medicine by major regulators such as the FDA or the EMA.
Clinical evidence comes mainly from small studies conducted in Russia, several of which were open-label or lacked robust blinding. Reported outcomes include lower anxiety scores, changes in attention measures, and effects on asthenic states following illness. Sample sizes are typically in the tens of participants, and independent replication outside the region is scarce. Reviews published in English generally note the limited methodological quality of the underlying trials. Whether the compound produces clinically meaningful effects under rigorous conditions remains unresolved.
Proposed mechanisms center on modulation of the GABA system, but no single molecular target has been confirmed. Rodent studies report changes in GABA-A receptor expression and in the turnover of serotonin, dopamine, and norepinephrine in several brain regions. Increases in brain-derived neurotrophic factor and its receptor have also been described after repeated administration. These findings come largely from animal models, and the degree to which they describe human neurochemistry remains an open question. The mechanism is best characterized as multi-system and not fully resolved.
Pharmacokinetic data are limited. Like most short peptides, Selank is vulnerable to plasma and tissue peptidases, and its measured half-life in circulation is short, on a minutes scale. The Pro-Gly-Pro tail slows this degradation but does not eliminate it. Intranasal administration is the route described in most reports, with absorption through the nasal mucosa and a hypothesized path into the central nervous system that avoids the blood-brain barrier. Direct measurements of human brain exposure are unavailable, so distribution claims rest on inference from animal work.
N0 is the initial quantity of the substance that will decay (this quantity may be measured in grams, moles, number of atoms, etc.), N(t) is the quantity that still remains and has not yet decayed after a time t, t½ is the half-life of the decaying quantity, τ is a positive number called the mean lifetime of the decaying quantity, λ is a positive number called the decay constant of the decaying quantity. The three parameters t½, τ, and λ are directly related in the following way:
=== Facilitates the quaternary structure === The enzyme cytochrome c oxidase, also known as Complex IV, is a large transmembrane protein complex found in mitochondria and bacteria. It is the last enzyme in the respiratory electron transport chain located in the inner mitochondrial or bacterial membrane. It receives an electron from each of four cytochrome c molecules, and transfers them to one oxygen molecule, converting molecular oxygen to two molecules of water. Complex IV has been shown to require two associated CL molecules in order to maintain its full enzymatic function. Cytochrome bc1 (Complex III) also needs cardiolipin to maintain its quaternary structure and functional role. Complex V of the oxidative phosphorylation machinery also displays high binding affinity for CL, binding four molecules of CL per molecule of complex V.
==== MPP salary increase and pension re-introduction ==== In 2025, Ford's government introduced legislation to grant MPPs a 35 per cent pay raise. Previously, an MPP earned $116,550 annually, which had not changed since 2009. With support from the other parties, the Legislature enacted the MPP Pension and Compensation Act, 2025, which increased MPPs' salaries to $157,350, ministers from $165,851 to $223,909, and the premier from $208,974 to $282,129. The change also re-introduced pensions for MPPs.
The NO can then contribute to two suggested pathways. Under typical conditions, nitric oxide (NO) interacts non-covalently with the heme and copper (Cu) subunits of CCO, competing with oxygen molecules and inhibiting cellular respiration, leading to reduced production of adenosine triphosphate (ATP). Low energy light can revise the mitochondrial inhibition of cellular respiration by photodissociating of NO from CCO, and thereby increasing ATP synthesis. The second pathway proposes that released nitric oxide (NO) will boost Cytochrome c Oxidase (CCO) activity, as an enzyme nitrite reductase. This mechanism includes the transfer of electrons to oxygen molecules, resulting in the production of water and reactive oxygen species (ROS). The ROS then activates enzymes necessary for producing vital cellular components like nucleic acids and proteins. LED therapy may also increase ROS, which may activate transcription factors that manage genes important for cell growth, cytokine production, and making growth factors for cell repair and proliferation.
On 15 May, an armed mob, headed by François-Vincent Raspail, Auguste Blanqui, and Armand Barbès, and assisted by the proletariat-aligned Guard, attempted to overwhelm the Assembly, but were defeated by the bourgeois-aligned battalions of the National Guard. Meanwhile, the national workshops were unable to provide remunerative work for the genuine unemployed, and of the thousands who applied, the greater number were employed in aimless digging and refilling of trenches; soon even this expedient failed, and those for whom work could not be invented were given a half wage of 1 franc a day. On 21 June, Alfred de Falloux decided in the name of the parliamentary commission on labour that the workmen should be discharged within three days and those who were able-bodied should be forced to enlist in the armed forces.

=== Reaction === The peptide to be sequenced is adsorbed onto a solid surface. One common substrate is glass fibre coated with polybrene, a cationic polymer. The Edman reagent, phenylisothiocyanate (PITC), is added to the adsorbed peptide, together with a mildly basic buffer solution of 12% trimethylamine. This reacts with the amine group of the N-terminal amino acid. The terminal amino acid can then be selectively detached by the addition of anhydrous acid. The derivative then isomerises to give a substituted phenylthiohydantoin, which can be washed off and identified by chromatography, and the cycle can be repeated. The efficiency of each step is about 98%, which allows about 50 amino acids to be reliably determined.
However, the details of how serpin polymers cause cell death remains to be fully understood. Physiological serpin polymers are thought to form via domain swapping events, where a segment of one serpin protein inserts into another. Domain-swaps occur when mutations or environmental factors interfere with the final stages of serpin folding to the native state, causing high-energy intermediates to misfold. Both dimer and trimer domain-swap structures have been solved. In the dimer (of antithrombin), the RCL and part of the A-sheet incorporates into the A-sheet of another serpin molecule. The domain-swapped trimer (of antitrypsin) forms via the exchange of an entirely different region of the structure, the B-sheet (with each molecule's RCL inserted into its own A-sheet). It has also been proposed that serpins may form domain-swaps by inserting the RCL of one protein into the A-sheet of another (A-sheet polymerisation). These domain-swapped dimer and trimer structures are thought to be the building blocks of the disease-causing polymer aggregates, but the exact mechanism is still unclear.
Richard Laurence Millington Synge (28 October 1914 – 18 August 1994) was an English biochemist, and shared the 1952 Nobel Prize in Chemistry for the invention of partition chromatography with Archer Martin.
No relapse of flystrike occurred. Mules and others soon developed this serendipitous discovery into a technique now known as mulesing. During this operation, small strips of epidermis are peeled from a sheep's buttock using steel shears on either side of the anus and underside of the tail. This was formerly performed on mature sheep but it was later found that lambs recover more quickly and completely than older animals. Mulesing reduces the likelihood of flystrike by about 13 times. The practice became nearly universal during the 20th century. The success of animal rights movements in agitating for the procedure's curtailment has brought the proportion of Australian sheep ranchers who practice mulesing down to around 70% today.

== Pathology == Abnormally elevated levels of glucagon may be caused by pancreatic tumors, such as glucagonoma, symptoms of which include necrolytic migratory erythema, reduced amino acids, and hyperglycemia. It may occur alone or in the context of multiple endocrine neoplasia type 1. Elevated glucagon is the main contributor to hyperglycemic ketoacidosis in undiagnosed or poorly treated type 1 diabetes. As the beta cells cease to function, insulin and pancreatic GABA are no longer present to suppress the freerunning output of glucagon. As a result, glucagon is released from the alpha cells at a maximum, causing a rapid breakdown of glycogen to glucose and fast ketogenesis. It was found that a subset of adults with type 1 diabetes took 8 hours longer on average (18 hours vs 10 hours) to approach ketoacidosis when given somatostatin (inhibits glucagon production) with no insulin. Inhibiting glucagon has been a popular idea of diabetes treatment, however, some have warned that doing so will give rise to brittle diabetes in patients with adequately stable blood glucose. The absence of alpha cells (and hence glucagon) is thought to be one of the main influences in the extreme volatility of blood glucose in the setting of a total pancreatectomy.
=== Adults === The World Health Organization recommends a minimal acceptable intake of approximately 1.3 mg/day. These values are considered to be adequate and safe for most of the general population. In North America, the U.S. Institute of Medicine (IOM) set the Recommended Dietary Allowance (RDA) for copper for healthy adult men and women at 0.9 mg/day. As for safety, the IOM also sets Tolerable upper intake levels (ULs) for vitamins and minerals when evidence is sufficient. In the case of copper, the UL is set at 10 mg/day. The European Food Safety Authority reviewed the same safety question and set its UL at 5 mg/day.
The war has severely weakened Yemen’s health system. More than half of the country’s health facilities have been reported as non-functional, a situation attributed to airstrikes, damage to buildings, departures or non-payment of staff, and long-term shortages of medicines and equipment.Facilities that continue to operate frequently do so under difficult conditions, facing unreliable electricity, scarce fuel for generators and obstacles in maintaining hygiene standards or cold-chain storage. Researchers link these operation difficulties to port restrictions, disrupted supply chains and the broader economic crisis. This institutional collapse has coincided with a series of major epidemics. Yemen has experienced what many researchers describe as the largest recorded cholera outbreak in modern times, with over one million suspected cases and earlier World Health Organization estimates of more than 600,000 cases with over 2,000 deaths. The spread of cholera has been associated with the destruction of water treatment plants, malfunctioning sewage systems and fuel shortages that hindered water pumping and waste disposal. Interruptions in vaccination campaigns and the wider deterioration of primary health care also facilitated outbreaks of diphtheria and measles, while when the COVID-19 pandemic reached Yemen, observers reported delayed vaccine delivery, limited testing capacity and additional economic pressure due to rising prices and economic stagnation.
=== Neuronal proliferation === NGF can drive the expression of genes such as bcl-2 by binding to the Tropomyosin receptor kinase A, which stimulates the proliferation and survival of the target neuron. High affinity binding between proNGF, sortilin, and p75NTR can result in either survival or programmed cell death (PCD). Study results indicate that superior cervical ganglia neurons that express both p75NTR and TrkA die when treated with proNGF, while NGF treatment of these same neurons results in survival and axonal growth. Survival and PCD mechanisms are mediated through adaptor protein binding to the death domain of the p75NTR cytoplasmic tail. Survival occurs when recruited cytoplasmic adaptor proteins facilitate signal transduction through tumor necrosis factor receptor members such as TRAF6, which results in the release of nuclear factor κB (NF-κB) transcription activator. NF-κB regulates nuclear gene transcription to promote cell survival. Alternatively, programmed cell death occurs when TRAF6 and neurotrophin receptor interacting factor (NRIF) are both recruited to activate c-Jun N-terminal kinase (JNK); which phosphorylates c-Jun. The activated transcription factor c-Jun regulates nuclear transcription via AP-1 to increase pro-apoptotic gene transcription.
=== Sleeping sickness treatment === The drug was registered for the treatment of gambiense sleeping sickness on 28 November 1990. However, in 1995 Aventis (now Sanofi-Aventis) stopped producing the drug, whose main market was African countries, because it did not make a profit. In 2001, Aventis and the WHO formed a five-year partnership, during which more than 320,000 vials of pentamidine, over 420,000 vials of melarsoprol, and over 200,000 bottles of eflornithine were produced by Aventis, to be given to the WHO and distributed by the association Médecins sans Frontières (also known as Doctors Without Borders) in countries where sleeping sickness is endemic. According to Médecins sans Frontières, this only happened after "years of international pressure," and coinciding with the period when media attention was generated because of the launch of another eflornithine-based product (Vaniqa, for the prevention of facial-hair in women), while its life-saving formulation (for sleeping sickness) was not being produced.

Identity is confirmed by matching the retention time in reversed-phase chromatography against a reference standard and by measuring the molecular mass with mass spectrometry. Tandem mass spectrometry or amino acid analysis can verify the sequence of the seven residues. Because the peptide contains no aromatic amino acids, detection at 280 nm is not useful.
Lyophilized powder is normally stored desiccated at -20 °C, protected from light and moisture. Powder kept under these conditions is generally stable for long periods. Solutions are less stable and are usually prepared immediately before use.
A purity value from one chromatographic method does not capture every possible impurity. Related peptides with similar retention behavior, counterions, and residual solvents may not appear in the same analysis. Independent testing with an orthogonal method provides stronger assurance of identity and content.
It is a seven-amino-acid peptide, Thr-Lys-Pro-Arg-Pro-Gly-Pro, produced by chemical synthesis rather than extracted from biological tissue. Its design is based on tuftsin, a natural immunomodulatory tetrapeptide. The C-terminal Pro-Gly-Pro segment is a common stabilising motif in short regulatory peptides.
