A practical reference on Semax: what it is, how it behaves, what the literature reports, and where the honest uncertainties sit.

Pharmacokinetic data are sparse and largely derived from animal work. After intranasal administration the peptide appears in plasma within minutes, and reported half-lives are short, on the order of minutes to tens of minutes. Degradation proceeds through ordinary proteolytic cleavage into constituent amino acids and smaller fragments. Direct evidence that intact Selank reaches brain tissue in meaningful amounts is limited, and the extent of blood-brain barrier penetration is debated. Some authors argue that fragments, not the parent peptide, carry much of the observed activity.
Published clinical work is concentrated in Russian-language journals and generally involves small samples without independent replication. Systematic reviews in English note the shortage of randomised, placebo-controlled trials and the difficulty of verifying methods from translated reports. Outcome measures vary between studies, which complicates pooling of results. Interest in the compound as a cognitive or anxiolytic agent therefore rests on a thinner evidence base than the volume of citations suggests. Replication in well-powered trials with preregistered endpoints would be needed before firm conclusions about efficacy can be drawn.
Proposed mechanisms centre on the GABAergic system. Animal and tissue studies report changes in GABA-A receptor expression and reduced activity of GABA transaminase, the enzyme that degrades GABA. Effects on monoamine turnover, including serotonin and dopamine pathways, are also described, and a separate line of work links the peptide to increased expression of brain-derived neurotrophic factor in hippocampal tissue. Most of these findings come from rodent models and cell preparations. How the individual observations combine into a single coherent mode of action is not settled.
Handling follows standard practice for research peptides. Material is weighed in a low-humidity environment because the powder absorbs atmospheric moisture. Purity is reported as the percentage area of the main peak in a chromatogram, with specifications commonly set at 95 percent or higher; values below that threshold indicate the presence of truncated or modified species. Residual trifluoroacetate from purification is often present and may affect mass balance. Certificates of analysis should state the analytical method, the column and gradient used, and the lot-specific retention time so that results can be compared across suppliers.
Identity and purity of selank are established with reversed-phase high-performance liquid chromatography coupled to mass spectrometry. The peptide elutes from C18 columns with acetonitrile gradients in water containing trifluoroacetic acid or formic acid, and detection is usually performed by ultraviolet absorbance near 214 nm. Electrospray ionization in positive mode gives a doubly protonated ion near m/z 377, consistent with a mass of about 752 Da. Amino acid analysis or tandem mass spectrometry of fragment ions confirms the sequence. Because the molecule has no aromatic residues, it lacks a usable 280 nm chromophore, so low-wavelength detection or mass spectrometry is required.
Peptide bonds in selank are susceptible to hydrolysis under strongly acidic or basic conditions, and the terminal proline residues are vulnerable to exopeptidase activity in biological samples. Lyophilized powder stored dry at -20 °C typically remains stable for extended periods, whereas aqueous solutions degrade faster and may lose measurable purity within days to weeks depending on pH, temperature, and microbial load. Repeated freeze-thaw cycles promote aggregation and adsorption to container surfaces. For analytical work, solutions are usually prepared fresh, kept cold, and used within a single working day.

Reported pharmacological effects center on reduced anxiety-like behavior in animal models and on measures of memory and learning. Proposed contributing mechanisms include modulation of GABAergic signaling, shifts in monoamine turnover, and changes in the activity of enzymes that degrade neuropeptides. Effects on the expression of genes linked to neuroplasticity have also been described. No single molecular target is widely accepted, and whether the behavioral findings arise from one pathway or several remains an open question.
Selank is a synthetic heptapeptide with the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro. It was designed as a stabilized analogue of tuftsin, a naturally occurring tetrapeptide fragment derived from the immunoglobulin heavy chain. The additional Pro-Gly-Pro segment at the carboxyl terminus is intended to slow enzymatic cleavage. The compound is usually described in the literature as a synthetic peptide with anxiolytic and cognitive-related activity, a label that reflects a research context rather than an approved therapeutic category.
Naming for this compound is not fully standardised in English sources. The spelling Selanc appears in some transliterations, and catalogue entries may instead list the peptide sequence itself as the identifier. Reference material sometimes groups it with other short synthetic peptides studied for behavioural effects, which can create confusion when citations are compared. Distinguishing the exact sequence from related tuftsin analogues is therefore a practical first step when reviewing any dataset or specification sheet.
Selank is a synthetic heptapeptide with the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro, frequently abbreviated as TKPRPGP. It was designed as a structural analogue of tuftsin, a naturally occurring tetrapeptide released by enzymatic cleavage of the immunoglobulin heavy chain. The two additional proline residues at the C-terminal end extend the parent chain and change how the molecule behaves in solution. The free peptide has a calculated molecular mass of approximately 751.9 g/mol and is generally supplied as a lyophilised white to off-white powder.
Development work on the compound began in the 1980s and 1990s at the Institute of Molecular Genetics in Moscow, within the same research programme that produced the peptide Semax. Early investigators sought a tuftsin derivative with improved resistance to enzymatic breakdown and with activity in the central nervous system after peripheral administration. Most of the primary literature from this period was published in Russian, a factor that still shapes how easily the findings can be checked by outside groups.
Once dissolved, the peptide is markedly less stable than the dry powder. Aqueous solutions are subject to backbone hydrolysis and to microbial growth when they are handled without sterile technique. Buffered solutions near neutral pH are common for short-term laboratory work, while acidic conditions are sometimes used to improve solubility. Analytical laboratories generally prepare working solutions fresh rather than storing them, and a residual water film left in a reopened vial can seed degradation even when the container appears dry.
Reversed-phase high-performance liquid chromatography is the standard technique for estimating peptide purity. The result is a peak-area percentage, which describes how much of the detected material elutes as the main peak in one run. Mass spectrometry confirms the molecular mass and can reveal truncated, adducted, or otherwise modified species. Amino acid analysis or tandem mass spectrometry can address sequence fidelity when identity is in doubt. None of these measurements, taken alone, establishes that a sample is fit for any specific purpose.
Lyophilized selank is normally supplied as a dry powder and is considered stable for extended periods when kept cold and dry. Moisture uptake is the main practical threat, because absorbed water promotes both hydrolysis and aggregation in the solid state. Vials are usually warmed to room temperature before opening so that condensation does not form on the powder. Supplier documentation commonly specifies -20 °C for routine storage, with -80 °C used for material intended to be archived for years.
== Transition from unicellularity to multicellularity == For the majority of Earth’s history life has been unicellular. However, unicellular organisms had the ingredients in them for multicellularity to arise; despite this, organisms were restricted due to the lack of hospitable environmental conditions. The rise of atmospheric oxygen (The Great Oxygenation Event, 2.46–2.06 bya) led organisms to be able to develop more complex body plans. In order for multicellularity to have occurred, organisms must have been capable of cellular communication, aggregation, and the formation of specialised tissues. The transition to multicellularity that began the evolution of animals from protozoa is one of the most poorly understood of history’s life events. Understanding choanoflagellates and their relation to sponges is important when positing theories on the origins of multicellularity
Axonotmesis are a range of peripheral nerve injuries that are considered more severe than Neurapraxia and less severe than Neurotmesis. Axonotmesis involves loss of relative axon continuity and myelin covering, but preservation of the connective tissue framework (including encapsulating tissue, the epineurium and perineurium). Since connective tissue framework is preserved then axonal regeneration is possible, but recovery is slower. Other characteristics:
Overtraining occurs when a bodybuilder has trained to the point where their workload exceeds their recovery capacity. There are many reasons why overtraining occurs, including lack of adequate nutrition, lack of recovery time between workouts, insufficient sleep, and training at a high intensity for too long (a lack of splitting apart workouts). Training at a high intensity too frequently also stimulates the central nervous system (CNS) and can result in a hyperadrenergic state that interferes with sleep patterns. To avoid overtraining, intense frequent training must be met with at least an equal amount of purposeful recovery. Timely provision of carbohydrates, proteins, and various micronutrients such as vitamins, minerals, phytochemicals, even nutritional supplements are critical. A mental disorder, informally called bigorexia (by analogy with anorexia), may account for overtraining in some individuals. Sufferers feel as if they are never big enough or muscular enough, which forces them to overtrain to try to reach their goal physique. An article by Muscle & Fitness magazine, "Overtrain for Big Gains", claimed that overtraining for a brief period can be beneficial. Overtraining can be used advantageously, as when a bodybuilder is purposely overtrained for a brief period of time to super compensate during a regeneration phase. These are known as "shock micro-cycles" and were a key training technique used by Soviet athletes.
Minute by Minute's triumph was bittersweet because it coincided with the dissolution of the band. The pressure of touring while recording and releasing an album each year had worn the members down. Baxter and McDonald had been in creative conflict for some time. McDonald desired a direct, soulful and polished rock/R&B sound, while Baxter insisted on embellishing guitar parts in an increasingly avant-garde style. (Both McDonald and Baxter elaborated on the matter in the documentary series Behind the Music, which aired on VH1 in February 2001.) The Doobie Brothers toured Japan in early 1979 to support Minute by Minute, but upon their return, the band broke up. Once Minute by Minute's success became apparent, they opted to reunite, but without Baxter, who McDonald felt he could no longer work with. Hartman and LaKind also left the band. (Hartman subsequently joined Johnston's touring band in 1979 and taped an appearance with him that aired on Soundstage in 1980.) Looking to capitalize on the momentum provided by the success of "Minute by Minute", the remaining Doobies (Simmons, Knudsen, McDonald and Porter) decided to embark on a national tour with a reconstituted lineup. In 1979 Hartman was replaced by session drummer Chet McCracken and Baxter by multi-instrumental string player John McFee (late of Huey Lewis's early band Clover); Cornelius Bumpus (who had been part of a recent reunion of Moby Grape) was also recruited to add vocals, keyboards, saxophones, and flute to the lineup.

Developments in Britain helped to alienate the white Rhodesian population from the former mother country, namely Queen Elizabeth II's 1966 speech to the Parliament of Jamaica where she endorsed majority rule and her attempt in 1968 to block the hanging of what Rhodesians perceived as Zimbabwean nationalist terrorists. Until late 1969, Rhodesia still recognised Queen Elizabeth II as head of state, even though it opposed the British government itself for hindering its goals of independence. However she refused to accept the title Queen of Rhodesia and eventually the Smith government abandoned their attempts to remain loyal to the British Crown. In a 1969 referendum, a majority of the electorate voted to declare Rhodesia an independent republic. The hope being that this move would facilitate recognition as an independent state by the international community, but the issue of white minority rule remained and continued to hinder this effort, and like the UDI before it, the proclamation of a republic lacked international recognition. The British public overall viewed the issue of Rhodesia and UDI as a distant and anachronistic one that they cared relatively little about and saw any expedient response to it by their government as satisfactory. Most Britons saw the end of Rhodesia's white supremacist timocracy as inevitable and justified. Although the majority Britons opposed military intervention against the renegade Rhodesian state, this opinion was not uniformly held.

Corticosteroids have become some of the most commonly incorporated lightening agents. Long-term usage over large areas of skin may promote percutaneous absorption, which can produce complications such as skin atrophy and fragility, glaucoma, cataracts, edemas, osteoporosis, menstrual irregularities, and growth suppression. A 2000 study performed in Dakar, Senegal, indicated that chronic usage of skin lighteners was a risk factor for hypertension and diabetes. Chemically lightened skin is more highly susceptible to sun damage and dermal infection. Long-term users of skin bleachers can easily develop fungal infections and viral warts. Pregnant users may also experience health complications for both them and their children.
Conversely, if the UK had such a device, the population could be protected. In 1934, along with a movement to establish a scientific committee to examine these new types of weapons, the RAF offered a £1,000 prize to anyone who could demonstrate a working model of a death ray that could kill a sheep at 100 yards; it went unclaimed.

=== Generating diagnostic hypotheses === Determining which pieces of information are most important to the next phase of the diagnostic process is of vital importance. It is during this stage that clinical bias like anchoring or premature closure may be introduced. Once key findings are determined, they are compared to profiles of possible diseases. These profiles include findings that are typically associated with the disease and are based on the likelihood that someone with the disease has a particular symptom. A list of potential diagnoses is termed the "differential diagnosis" for the patient and is typically ordered from most likely to least likely, with special attention given to those conditions that have dire consequences for the patient if they were missed. Epidemiology and endemic conditions are also considered in creating and evaluating the list of diagnoses. The list is dynamic and changes as the doctor obtains additional information that makes a condition more ("rule-in") or less ("rule-out") likely based on the disease profile. The list is used to determine what information will be acquired next, including which diagnostic test or imaging modality to order. The selection of tests is also based on the doctor's knowledge of the specificity and sensitivity of a particular test. An important part of this process is knowledge of the various ways that a disease can present in a patient. This knowledge is gathered and shared to add to the database of disease profiles used by doctors. This is especially important in rare diseases.
Shortly before the war, Hans von Halban and Lew Kowarski moved their research on neutron moderation from France to Britain, smuggling the entire global supply of heavy water (which had been made in Norway) across in twenty-six steel drums. During World War II, Nazi Germany was known to be conducting experiments using heavy water as moderator for a nuclear reactor design. Such experiments were a source of concern because they might allow them to produce plutonium for an atomic bomb. Ultimately it led to the Allied operation called the "Norwegian heavy water sabotage", the purpose of which was to destroy the Vemork deuterium production/enrichment facility in Norway. At the time this was considered important to the potential progress of the war. After World War II ended, the Allies discovered that Germany was not putting as much serious effort into the program as had been previously thought. The Germans had completed only a small, partly built experimental reactor (which had been hidden away) and had been unable to sustain a chain reaction. By the end of the war, the Germans did not even have a fifth of the amount of heavy water needed to run the reactor, partially due to the Norwegian heavy water sabotage operation. However, even if the Germans had succeeded in getting a reactor operational (as the U.S. did with Chicago Pile-1 in late 1942), they would still have been at least several years away from the development of an atomic bomb.
Granola bars are made of granola, muesli or cereal, typically including oat flakes, vegetable fats or oils, and some type of sweetener. Many granola bars contain additional ingredients such as nuts, dried fruits, and seeds. They become popular in the 1960s as part of the counterculture Hippie movement.
Register and use state prescription drug monitoring programs Enhance education and training Support comprehensive treatment for pain and substance use disorders Help end stigma Co-prescribe naloxone to patients at risk of overdose Encourage safe storage and disposal of opioids and all medications. The Opioid Task Force 2018 Progress Report states that between 2013 and 2017 opioid prescriptions have decreased by 22.2%, which includes a 9% decrease from 2016 to 2017 alone. The AMA Opioid Task Force also reports a 389% increase in physician participation in PDMPs. Further, physicians are encouraged to co-prescribe naloxone to those at risk of overdose. In 2017 alone, weekly filled naloxone prescriptions have more than doubled from 3,500 to 8,000 and more than 50,000 physicians were certified in 2017 to provide in-office buprenorphine. Patrice A. Harris, chair of the AMA Opioid Task Force, urges increased participation by physicians, saying "what is needed now is a concerted effort to greatly expand access to high quality care for pain and for substance use disorders. Unless and until we do that, this epidemic will not end."
The evidence for the effectiveness of non-pharmacological interventions for chronic pain is insufficient to recommend such interventions alone, however their use in combination with medications may be reasonable. There is some evidence that aquatic therapy is a beneficial intervention. The spasticity associated with MS can be difficult to manage because of the progressive and fluctuating course of the disease. Although there is no firm conclusion on the efficacy in reducing spasticity, PT interventions can be a safe and beneficial option for patients with multiple sclerosis. Physical therapy including vibration interventions, electrical stimulation, exercise therapy, standing therapy, and radial shock wave therapy (RSWT), were beneficial for limiting spasticity, helping limit excitability, or increasing range of motion.

Reports describe modulation of GABA signalling, changes in monoamine turnover and effects on neurotrophic factor expression. These observations come mainly from animal and cell studies. A single unifying mechanism has not been demonstrated.
The peptide enters plasma rapidly and is broken down by ordinary proteases into amino acids and shorter fragments. Reported half-lives are short. Whether meaningful amounts of the intact molecule reach the brain is an open question.
Most clinical reports are small, published in Russian and not independently replicated. English-language reviews highlight the absence of large randomised trials. Conclusions about efficacy should be treated as provisional.
Identity is confirmed by matching the retention time in reversed-phase chromatography against a reference standard and by measuring the molecular mass with mass spectrometry. Tandem mass spectrometry or amino acid analysis can verify the sequence of the seven residues. Because the peptide contains no aromatic amino acids, detection at 280 nm is not useful.
