=== Treatment of deficiency === Severe vitamin B12 deficiency is initially corrected with daily intramuscular injections of 1000 μg of the vitamin, followed by maintenance via monthly injections of the same amount or daily oral dosing of 1000 μg. The oral daily dose far exceeds the vitamin requirement because the normal transporter protein-mediated absorption is absent, leaving only inefficient intestinal passive absorption. Injection side effects include skin rash, itching, chills, fever, hot flushes, nausea and dizziness. == Pathology == Some metabolic myopathies involve the under- or over-utilization of the purine nucleotide cycle. Metabolic myopathies cause a low ATP reservoir in muscle cells (ADP > ATP), resulting in exercise-induced excessive AMP buildup in muscle, and subsequent exercise-induced hyperuricemia (myogenic hyperuricemia) through conversion of excessive AMP into uric acid by way of either AMP → adenosine or AMP → IMP. During strenuous exercise, AMP is created through the use of the adenylate kinase (myokinase) reaction after the phosphagen system has been depleted of creatine phosphate and not enough ATP is being produced yet by other pathways (see above reaction in 'Occurrence' section). In those affected by metabolic myopathies, exercise that normally wouldn't be considered strenuous for healthy people, is however strenuous for them due to their low ATP reservoir in muscle cells. This results in regular use of the myokinase reaction for normal, everyday activities. Besides the myokinase reaction, a high ATP consumption and low ATP reservoir also increases protein catabolism and salvage of IMP, which results in increased AMP and IMP. These two nucleotides can then enter the purine nucleotide cycle to produce fumarate which will then produce ATP by oxidative phosphorylation. If the purine nucleotide cycle is blocked (such as AMP deaminase deficiency) or if exercise is stopped and increased fumarate production is no longer needed, then the excess nucleotides will be converted into uric acid.

== Applications == HBsAg made through recombinant DNA is used to make the hepatitis B vaccine. It has a very good efficacy of about 95%, with protection lasting for more than 30 years, even after the anti-HbsAg antigen titers have fallen. The RTS,S also makes use of HBsAg. It is a mixture of a version of malaria surface antigen grafted to HBsAg (RTS) and ordinary HBsAg (S), both made through recombinant DNA. Much like ordinary HBsAg, these two are able to assemble into virus-like particles that are soluble in water.
== Side effects == Like any form of radiotherapy, ionising radiation can harm healthy tissue as well as the intended treatment target. Radiation from lutetium (177Lu) oxodotreotide can cause damage when the medicine passes through tubules in the kidney. Arginine/lysine can be used to reduce renal radiation exposure during peptide receptor radionuclide therapy with lutetium (177Lu) oxodotreotide.

== Post-World-War-II life and work == In 1947, aged 31, Crick began studying biology and became part of an important migration of physical scientists into biological research. This migration was made possible by the newly won influence of physicists such as Sir John Randall, who had helped win the war with inventions such as radar. Crick had to adjust from the "elegance and deep simplicity" of physics to the "elaborate chemical mechanisms that natural selection had evolved over billions of years." === Valve Corporation v. Rothschild === In 2015, Display Technologies LLC, one of several IP-holding companies owned by Leigh Rothschild, sued Valve over patent infringement. While the suit was dismissed by January 2016, Valve shortly followed by acquiring licenses to twenty patents owned across Rothschild's firms. Around 2022, another Rothschild company, Patent Asset Management, began sending Valve demands to buy new licenses to two of the patents Valve already had licensed. Valve filed a lawsuit against the company in 2023 under Washington state's Patent Troll Protection and Consumer Protection Acts. In the months that followed, at least three lawsuits were filed in Texas by Rothschild's companies against Valve accusing Valve of additional patent infringement. Valve argued these suits were retaliatory to their Washington suit, and requested them to be dismissed. In February 2026, a jury found in Valve's favor on all counts.
==== First-pass effect and differences from other routes ==== The first-pass effect that occurs with oral estradiol results in unusually high levels of estrone and estrogen conjugates in the circulation as well as of estradiol in the liver. These unique properties of oral estradiol result in a number of pharmacological differences relative to the other routes of administration of estradiol. The high levels of estrone and estrogen conjugates that occur with oral estradiol raise the question of the pharmacodynamic significance of these metabolites. In contrast to estradiol however, estrone has very low activity as an estrogen. The affinities of estrone for the human ERs and its estrogenic activity have been reported to be approximately 3 to 4% of those of estradiol. In addition, unlike estradiol and estriol, estrone is not accumulated in target tissues. Because estrone can be transformed into estradiol, most of its activity in vivo is actually due to conversion into estradiol. In accordance, doses of oral and transdermal estradiol that achieve similar levels of estradiol have been found, in spite of markedly elevated levels of estrone with oral estradiol but not with transdermal estradiol, to possess equivalent and non-significantly different potency in terms of clinical measures including suppression of LH and FSH levels, inhibition of bone resorption, and relief of menopausal symptoms such as hot flashes.
