== Procedure == Restriction enzymes are used to excise the gene of interest (the insert) from the parent. The insert is purified in order to isolate it from other DNA molecules. A common purification method is gel isolation. The number of copies of the gene is then amplified using polymerase chain reaction (PCR). Simultaneously, the same restriction enzymes are used to digest (cut) the destination. The idea behind using the same restriction enzymes is to create complementary sticky ends, which will facilitate ligation later on. A phosphatase, commonly calf-intestinal alkaline phosphatase (CIAP), is also added to prevent self-ligation of the destination vector. The digested destination vector is isolated/purified. The insert and the destination vector are then mixed together with DNA ligase. A typical molar ratio of insert genes to destination vectors is 3:1; by increasing the insert concentration, self-ligation is further decreased. After letting the reaction mixture sit for a set amount of time at a specific temperature (dependent upon the size of the strands being ligated; for more information see DNA ligase), the insert should become successfully incorporated into the destination plasmid.

== Mechanism of toxicity == Once azinphos-methyl is absorbed it can cause neurotoxic effects, like other organophosphate insecticides. At high concentrations AzM itself can be toxic because it can function as an acetylcholinesterase (AChE) inhibitor. But its toxicity is mainly due to the bioactivation by a cytochrome P450 (CYP450)-mediated desulfuration to its phosphate triester or oxon(gutoxon) (see figure 2). Gutoxon can react with a serine hydroxyl group at the active site of the AChE. The active site is then blocked and AChE is inactivated. Under normal circumstances acetylcholinesterase rapidly and efficiently degrades the neurotransmitter acetylcholine (ACh) and thereby terminates the biological activity of acetylcholine. Inhibition of AChE results in an immediate accumulation of free unbound ACh at the ending of all cholinergic nerves, which leads to overstimulation of the nervous system.

== Career == After his studies at Leipzig had ended, he began a year of work at the Moabit Hospital in Berlin as the resident pathologist. During this time, he began his research and studies on the issue of atherosclerosis. His first published works, dated during this period were in on the development of atherosclerosis in experimental animals when administered cholesterol. He also spent time working in the laboratory of Peter Rona at the Berlin Municipal Hospital. In 1926, Schoenheimer was invited by Ludwig Aschoff to join the faculty at the University of Freiburg. There he worked as an assistant professor. The investigation of pathological material was part of his work duties. During this time, he also researched atherosclerosis and the role of dietary cholesterol in its development. He became the active in 1927, and then the titular, head of his division in 1931. In 1930, until 1931, Schoenheimer was in the United States as the Douglas Smith Fellow at the University of Chicago. During this time Schoenheimer came into contact with the Josiah Macy Jr Foundation. Later in 1931, The Macy Foundation, with Ludwig Kast as its president, started supporting Schoenheimer in his atherosclerosis studies. Following his fellowship, he returned to the University of Freiburg, taking the position of head of the Pathological Chemistry department.
