== History == The chemical elements came into being in two phases. The first commenced shortly after the Big Bang. The monocyte monolayer assay (MMA) is used to determine the clinical significance of alloantibodies produced by blood transfusion recipients. The assay is used to assess the potential for intravascular hemolysis when incompatible cellular blood products are transfused to the anemic patient. When donor cells possess substances that are not produced by the recipient, the recipient's immune system produces antibodies against the substance; these are called alloantibodies. Specific white blood cells, called monocytes, are tasked with ingesting foreign material and become activated during certain inflammatory events. These activated monocytes come in contact with antibody-sensitized red blood cells (RBC) and may or may not exhibit phagocytosis (ingestion) and destroy the donor red blood cells. If monocytes destroy the RBC, the antibody attached to those RBC is considered clinically significant.

== Detection techniques == Early methods to detect alpha-amanitin included thin-layer chromatography (TLC). In most solvent systems used in TLC, alpha-amanitin and beta-amanitin would travel at different rates, thus allowing individual identification of each toxin. Another early method was the Meixner test (also known as the Wieland test), which would detect amatoxins, but also yielded false positives for some compounds, such as psilocin. Capillary zone electrophoresis was also developed, but was not adequately sensitive for clinical samples, but sufficient for mushroom extracts. More recently, the use of high-performance liquid chromatography (HPLC) has become the preferred method, which allows for better resolution, reproducibility, and higher sensitivity. A range of detectors can be paired with HPLC, such as UV or mass spectrometry. As early as the 1980s, antibody-based assays (immunoassays) were developed for amanitin (but more often recognize amatoxins as the antibodies cross-react with some of the congeners). The earliest immunoassays were radioimmunoassays and then enzyme linked immunosorbent assays (ELISAs). More, recently, in 2020, a monoclonal antibody-based lateral flow immunoassay (similar to a pregnancy test) has been developed that can quickly and selectively detect amatoxins in mushrooms and in urine samples.
== Indications == The main effects of paregoric are to increase the muscular tone of the intestine, to inhibit normal peristalsis, and as an expectorant; a peer-reviewed clinical study in 1944 reported "that all of [its] ingredients have been found to contribute toward the expectorant action of paregoric, and, further, that an advantage is contained in the combination over the sum of the effects of the individual constituents," that Paregoric "is expectorant by virtue of a reflex from the stomach," and "preparations of paregoric which have aged for two or three years are superior as an expectorant to preparations aged for less time.". Its main medical use is to control fulminant diarrhea, and as an antitussive (cough suppressant). Problems with its use include opiate dependency and analgesia which can mask symptoms of diseases that need treatment. However, paregoric was characterized as "a needlessly complex pharmacopeial mixture... of a former day" by a 1966 study. In the 21st century its two main uses have been largely supplanted by minimally psychoactive cough-suppressant drugs (such as dextromethorphan) and non-psychoactive antidiarrheal drugs (such as loperamide).
An adenylating enzyme (Ad) catalyzes the condensation of decanoic acid and the N-terminal tryptophan, which incorporates decanoic acid into the growing peptide (Figure 3). Elongation by these NRPS modules ultimately leads to macrocyclization and release in which an α-amino group, namely threonine, acts as an internal nucleophile during cyclization to yield the 10-amino-acid ring (Figure 6). The termination module in the NRPS assembly line has a C-A-T-TE organization. The thioesterase domain catalyzes chain termination and release of the mature lipopeptide. The molecular engineering of daptomycin, the only marketed acidic lipopeptide antibiotic to date (Figure 8), has seen many advances since its inception into clinical medicine in 2003.
=== Branch-chain fatty acid synthase === This system functions similarly to the branch-chain fatty acid synthesizing system, however it uses short-chain carboxylic acids as primers instead of alpha-keto acids. In general, this method is used by bacteria that do not have the ability to perform the branch-chain fatty acid system using alpha-keto primers. Typical short-chain primers include isovalerate, isobutyrate, and 2-methyl butyrate. === Ligand-based === Ligand-based drug design (or indirect drug design) relies on knowledge of other molecules that bind to the biological target of interest. These other molecules may be used to derive a pharmacophore model that defines the minimum necessary structural characteristics a molecule must possess in order to bind to the target. A model of the biological target may be built based on the knowledge of what binds to it, and this model in turn may be used to design new molecular entities that interact with the target. Alternatively, a quantitative structure-activity relationship (QSAR), in which a correlation between calculated properties of molecules and their experimentally determined biological activity, may be derived. These QSAR relationships in turn may be used to predict the activity of new analogs.
== Selection rules == For any given atom, there are quantum numbers that can specify the wavefunction of that atom. Using the hydrogen atom as an example, four quantum numbers are required to fully describe the state of the system. Quantum numbers that are eigenvalues of the operators that commute with the wavefunction to describe physical aspects of the system, and are called “good” numbers because of this. Once good quantum numbers have been found for a given atomic transition, the selection rules determine what changes in quantum numbers are allowed. The electric dipole (E1) transition of a hydrogen atom can be described with the quantum numbers l (orbital angular momentum quantum number), ml (magnetic quantum number), ms (electron spin quantum number), and n (principal quantum number). When evaluating the effect of the electric dipole moment operator μ on the wavefunction of the system, we see that all values of the eigenvalue are 0, except for when the changes in the quantum numbers follow a specific pattern.
